Abstract:Conclusions: Common SNPs in the HFEgene influence susceptibility to HAND, particularly symptomatic impairment. These associations may indicate that iron dysregulation is most important in the mechanism(s) underlying progression from asymptomatic to symptomatic neurocognitive impairment. Associations with structural abnormalities in the brain such as brain atrophy, synaptodendriticloss, and abetadeposition are likely to be exacerbated by age, but further studies that oversample older adults are needed to determine interactions with age and/or age-specific effects.
